What a CAR-T potency assay measures

A CAR-T potency assay quantifies whether an engineered T cell product does what it was designed to do: recognize its target antigen, activate, and kill. Common readouts include target cell cytotoxicity, activation marker expression such as CD69 and CD25, cytokine release, and the proportion of CAR-positive cells in the product.

Every one of those readouts is measured against a target. If the target changes between runs, the potency number changes with it, and you have no way to tell whether the shift came from your product or your reagent. Antigen density on the target cell drives activation strength directly, which makes it the variable worth controlling first.

Fixed antigen density

Antigen density drives activation strength. Tumor cell lines drift with passage number, including antigen expression levels, so a line that expressed a marker at one density in year one will not match it in year three. A manufactured target holds its specification.

No donor sourcing

Primary cells vary between donors, do not scale, and bring consent and chain-of-custody considerations that get harder as a program moves toward commercial supply.

36-month shelf life, no cold chain

Non-biohazardous and shipped without dry ice. In-house cryopreserved CAR-T controls have been validated with a post-thaw working window of roughly six hours, which constrains how you schedule a run.

CAR-T potency assay FAQs

What is a CAR-T potency assay?

A test that measures whether a CAR-T product performs its intended biological function: recognizing its target antigen, activating, and killing target cells. Potency is a critical quality attribute and a required release test for licensed products.

Why use synthetic target cells instead of a tumor cell line?

Cell lines drift with passage number, so antigen density shifts over a program's lifetime. Since potency is expressed relative to that target, drift in the target propagates into the potency number. They also need continuous culture, banking and QC.

Do these controls require cold-chain shipping or biosafety handling?

No. StimCytes are non-biohazardous synthetic material with a 36-month shelf life and ship without dry ice. No pathogen screening or donor consent documentation.
